Effect of cis-Clopenthixol and Clopenthixol on the Swimming Ability and Locomotive Activity in Rats



Abstract
After cis-clopenthixol, clopenthixol and clebopride, known as dopamine receptor antagonist, were administered to normal rats, the alterations of the swimming ability and locomotive activity were determined. And the swimming abilities after administration of these drugs with apomorphine or lisuride to reserpinized rats were measured.
The results were as follow ;
1. When cis-clopenthixol(0.01-1.0§· / §¸, s.c.) and clopenthixol(0.01-1.0§· / §¸, s.c.) were administered to normal rats, the swimming abilities were significantly decreased against control rats dose-dependently.
2. When cis-clopenthixol, clopenthixol and clebopride(0.5§· / §¸, s.c.) were in jected to normal rats, the locomotive activities were markedly decreased in rats. But these effects were not observed in clopenthixol 0.01§· / §¸ -treated rats.
3. When given the same dose of two drugs to normal rats, cis-clopenthixol was more potent than clopenthixol.
4. When apomorphine(0.25§· / §¸, s.c.) and lisuride(0.2§· / §¸, s.c.) were administered to reserpinized rats, respectively, the swimming abilities were markedly increased in reserpinized rats.
5. When cis-clopenthixol(0.1§· / §¸, s.c.) and clebopride(0.5§· / §¸, s.c.) were injected with apomorphine 0.25§· / §¸ and lisuride 0.2§· / §¸, respectively, the swimming abilities were significantly reduced than that of apomorphine or lisuride alone. But the group of clopenthixol(0.1§· / §¸, s.c.) was not significant.
6. When compared with the effects of cis-clopenthixol and clopenthixol on the apomorphine or lisuride-induced hypermotility, cis-clopenthixol was shown to be more potent than clopenthixol.
